ABSTRACT
Soybean as the main food that contains vegetable protein sources such as soybean from year to year is increasing. Domestic soybean production has not been able to meet domestic needs so that soybean imports are taken. The high level of soybean import activities can pose a great risk for the spread of diseases, especially seed borne diseases, namely Peronosrica, an obligate parasitic fungus that can cause downy mildew disease in soybeans. Detection of the presence of P. manshurica in imported soybeans entering several regions in Sulawesi is an important step in preventing the entry and spread of Quarantine Plant Disturbing Organisms (QPTK) in Sulawesi. This study aimed to detect P. manshurica fungus in soybean imports in Surabaya and Sulawesi (Palu, Luwuk, Gorontalo, Kendari) using Polymerase Chain Reaction method. The analysis was carried out using a pair of specific primers PM3/PM4 comparing the presence or absence of DNA bands formed on agarose gel marked based on the thickness and clarity of DNA bands on amplicons visualization results measuring 283 bp with several test samples from Surabaya and Sulawesi (Palu, Luwuk, Gorontalo, Kendari). The results of morphological observations of all test samples have the same characteristics as the symptoms caused by the fungus P. mansuricha the symptoms of white to grayish crusts on the surface of infected soybeans with varying degrees of infection, the crusts are rough and dry to the touch and not easily separated from the seeds. These results are evidenced by molecular analysis using a pair of specific primers PM3/PM4 used for detection of the presence of P. manshurica in soybean showed all test samples were positive (+) for feather dew disease caused by the fungus P. mansuricha.
